showed no respiration due to endogenous substrates. Only L-ascorbate with NNN‘N’tetramethyl-p-phenylenediamine was oxidized by membrane particles and the specific activity obtained was lower than in corresponding particles prepared from NH4+-grown cells. Low-temperature reduced-minus-oxidized difference spectra (Fig. 1, trace b) suggested that this lack of electron-transport activity may be due, in part, to a marked decrease in synthesis of several cytochromes.

Characterization of Escherichia coli flagellar mutants that are insensitive to catabolite repression.
In Escherichia coli, the synthesis of the flagellar organelle is sensitive to catabolite repression. Synthesis requires the presence of the cyclic adenosine monophosphate receptor protein (Crp) and 3',5'-cyclic adenosine monophosphate (cAMP); i.e., mutants that lack Crp or adenylcyclase (Cya) synthesize no flagella. We isolated and characterized a series of mutants (cfs) that restored flagella-...
متن کاملCyclic adenosine monophosphate-independent mutants of the lactose operon of Escherichia coli.
In Escherichia coli the transcription of the lactose operon, like other catabolite-sensitive operons, requires catabolite gene activator protein and 3',5'-cyclic adenosine monophosphate in addition to ribonucleic acid polymerase. We isolated and analyzed lac(+) revertants from a crp(-) strain of E. coli. We found that revertants with a higher level of expression only for the lac operon lie in t...
متن کاملCharacterization of Escherichia coli adenylate cyclase mutants with modified regulation.
In Escherichia coli there is a large increase of cAMP synthesis in crp strains, which are deficient in the catabolite gene activator protein. In this work it was shown that this increase in cAMP synthesis does not occur in crp crr strains, deficient in both the catabolite gene activator protein and enzymeIII-glucose, a component of the phosphotransferase system. It was also shown that the other...

متن کاملEscherichia coli catabolite gene activator protein mutants defective in positive control of lac operon transcription.
We isolated three Escherichia coli catabolite gene activator protein mutants that are defective in the positive control of transcription initiation from the lac operon promoter region yet retain negative control of transcription from other promoters. One mutant has a substitution of valine for glutamate at residue 72, which lies in the cyclic AMP binding domain and contacts cyclic AMP. The othe...
